The original script for the sequel was supposed to be based on lawyers, as a follow up to the doctor story in Munna Bhai M.B.B.S. (2003). Rajkumar Hirani was not happy with the script and continued working for 2 years to develop the perfect sequel. A few scenes involving lawyers were nevertheless incorporated in this movie.
The scene in front of Mahatma Gandhi Library, Munna (Sanjay Dutt) and Circuit (Arshad Warsi) discuss about another radio jockey named Anupama Chopra who talks a lot about Shah Rukh Khan. In real life Anupama Chopra is a famous journalist, film critic. Anupama Chopra is also wife of the film's producer Vidhu Vinod Chopra and is the author of the book "King of Bollywood : Shah Rukh Khan and the seductive world of Indian cinema".
The term Gandhigiri became quite famous and people stated giving red roses to the corrupt official's so they could realize their mistakes.
Dilip Prabhawalkar was traveling by a tourist bus to Pune, and when he saw that a pirated version of the film was being screened on the bus while the film was still in theaters he immediately stopped the screening and informed the cops.

The radio jockey in the movie was modeled after real-life radio jockey Mallishka. Lead actress Vidya Balan spent time with Mallishka, who gained popularity with the now defunct Win 94.3 FM channel in Mumbai, in order to get the right inflection and attitude of a radio jockey.
The plot of the film was similar to Sanjeev Kumar starrer Yehi Hai Zindagi (1977) where Sanjeev Kumar happens to see God.
Indian Politicians were creating many issue regarding the term Gandhigiri which they felt was indecent.
